目的 采用高效液相色谱(HPLC)法和液质联用技术(UHPLC-MS)测定巴洛沙星片中的有关物质,并对大于0.1%的未知杂质进行定性分析。方法 采用Ecosil C18色谱柱(250 mm×4.6 mm,5 μm),以0.05 mol ·L-1磷酸二氢钾溶液(含0.2%三乙胺,用磷酸调节pH值为3.0)为流动相A,乙腈为流动相B,梯度洗脱,流速为1 mL ·min-1,检测波长294 nm。并采用UHPLC-MS技术对未知杂质进行了定性分析。结果 采用主成分自身稀释方法测定有关物质,方法的专属性强;采用液质联用技术对未知杂质进行了结构分析,判断杂质归属。结论 本法可简便、准确测定巴洛沙星片中的有关物质。 |
OBJECTIVE To use HPLC and UHPLC-MS method to determine and identify the related substances in Balofloxacin tablets,and determine qualitatively the unknown impurity greater than 0.1%. METHODS The Ecosil C18was used as the column.The mobile phase consisted of mobile phase A:0.05 mol ·L-1 potassium dihydrogen phosphate solution (containing 0.2% triethylamine,pH value was adjusted to 3.0 with phosphoric acid),mobile phase B:acetonitrile. Gradient elution chromatography was applied for the determination.The flow rate was 1 mL ·min-1,detection wavelength was at 294 nm.The unkown related substances were identified by UHPLC-MS.The structure of the unknown impurities was analyzed by UHPLC-MS. RESULTS Main component self-comparison could be used for testing related substances. The method was sensitive and specific. CONCLUSION The method is simple,sensitive and rapid,and it can be used for the determination of related substances in Balofloxacin tablets. |
